You can pray it any time of the day .. but people prefer it after salat al ishaa and before you sleep (before witir ofcourse).

Pray two Rakaa with the niya for salat al istikhara and as raed mentioned, think objectivly while praying .. and try to leave your heart aside ..
and then say this Duaa:

دعاء الاستخارة أو صلاة الاستخارة كما تسمى فهي : قال الرسول صلى الله عليه وسلم :" إذا هم أحدكم بالأمر فليركع ركعتين من غير الفريضة ثم ليقل : اللهم اني استخيرك بعلمك واستقدرك بقدرتك واسالك من فضلك العظيم فانك تقدر ولا اقدر وتعلم ولا اعلم وانت علام الغيوب ، اللهم إن كنت تعلم أن هذا الامر خير لي في ديني ومعاشي وعاقبة امري فاقدره لي ويسره لي ثم بارك لي فيه ، وان كنت تعلم ان هذا الامر شر لي في ديني ومعاشي وعاقبة امري فاصرفه عني واقدر لي الخير حيث كان ثم رضّني به ". ويسمي حاجته بدل كلمة الامر
